In a perfect world, no. A perfect screw will be perfectly positioned by a perfect thread. So, its simple - make them both perfect!
Seriously, in the real world there are manufacturing imperfections and clearances involved. The final position of a screw in a threaded hole is affected by many factors so the best you can say is that it is "close". That's why it is NEVER a good idea to try to use screw threads for any kind of precise positioning or alignment.
